In non-clinical studies, VELCADE has demonstrated⁵:

–	Inhibition of the cell cycle by stabilizing p21, p27, and p53
–	Inhibition of NF-ΚB pathway by stabilizing the NF-ΚB inhibitor, IΚB
–	Induction of apoptosis by activating pro-apoptotic Bcl-2 family members such as Bak2⁶

Reduced p27 protein levels correlate with shorter survival in MCL3⁷

–	Loss of p27 may be a result of protein degradation activity via the proteosome pathway
